Confusing sample project discovery.

At first glace I looked at this sample and was ready to hit "Cytoplasm".... however I can clearly see a staining over the blue portion.... in fact the staining is all over the cell and outside the red ....

So this one is tricky, it looks like a cell membrane, and cytoplasm and has staining inside the nucleus???

Right now I'm leaning toward unidentifiable....

I tried the tutorial and couldn't even figure that out. Just closed the window. I have no freaking clue what I'm supposed to do for sure or how the damn interface even works and I am far from a simpleton.

Maybe im over thinking it and making it hard because it's too simple or something.

Check the blue marker for holes, do those holes overlap with holes in the green as well? might be nucleoplasm + cytoplasm. Entire cell also sounds like plasma membrane but no spikes... show us blue only and green only

233: You pick what option(s) matches the picture on the left from the options on the right, you can zoom in to your sample and you can include/exclude colours with the r/b/g buttons to get a better view

From what I understand the green channel is what you are trying to guess. Blue and Red are simply overlays to help you better identify the structures. Its not easy at all. It can be confusing even for a human nevermind a computer.

Not cytoplasm according to the description, cyto is everywhere BUT the nucleus... unless it has both? Not sure.... like i said this one stumped me I'm almost ready to hit "unknown"

Well, the various components of the cell are either in the nucleus or outside of it, in the cell body, so if you get green staining in the nucleus and green staining outside of it, treat them as separate.

I usually ignore the body and toggle the green and blue channels to figure out which nucleus options are stained. Then I leave the blue channel on and ignore the nucleus, toggling red and green to figure out what's in the cell body. IMO, you're looking for dots vs. squiggles (Golgi or mitochondria) vs. spread-out watercolor (cytoplasm is a liquid and should look like watercolor).

Also, if there is something that is super-bright green, then I ignore the dark green background stuff.

EDIT: So in your image, I'd say nucleoplasm with some nuclear speckles that are extra bright, and those look like squiggles in the cell body but I can't see the whole image; zoom in and look around see if they're like this or like this.

Thanks Memphis Baas! Those are some really good tips. I agree about ignoring dark green if there are super-bright green features.

Tisiphone Dira is right, in the game you can have more than one correct answer. In this case it looks like nucleoplasm (green overlapping blue with holes corresponding to holes in the blue) and cytoplasm (general green in the cell body with no real identifiable features other than pretty uniformly green) to me.

I would say it's primarily in the nucleoplasm with a smaller portion in cytoplasm, not PM. There is no evidence concurrent with a specular nuclear pattern or staining of specific cytoplasmic organelles.

I'd go for nucleoplasm + cytoplasm.

Maybe with "cell-to-cell variations" too.
