I studied Latin. There is no combination of word stem and ending that will produce such a word. Given the entire word "Orthrus", clearly Orthr is just the word stem.

Orthrus is a an anglisized version of the Greek Orthros, which is of course a proper noun. It also is a certain genus of spider. But given CCP's inclination to name ships after gods/etc, the mythological creature is clearly the right choice.

Plural nominative 2nd declension masculine Greek nouns end in -oi. So Orthroi is my best guess.

Disclaimer: I did not study Greek. But learning Latin taught me which relevant key-words to google, "Greek noun declensions" in this case, when I want to know similar things about another language.
