I do 16 missions and get a storyline. But instead of doing the storyline I continue doing missions (because its a fleet op and I do not want to interrupt the flow to go haul 40,000 cu m of cargo). Will the missions I do count to the next storyline? Or do I need to do the offered storyline first? Can I get two or more storylines offered at once, all from doing missions for the same agent?

Yes. No. Yes.

Note that the next storyline will come from a different (the next closest) SL agent. This way you can rack up several SLs over the course of a week, although the last one will be several systems away.

Few days ago had 6 different Storyline missions in My Journal:
was doing standart missions from one Distribution agent... So Yes, multiple story ones...
