Depends.

In Faction Warfare, "plexing" refers to the act of going into a limited instance and orbiting a "button" of sorts until it times down. Once the timer reaches zero a person is awarded a certain amount of Loyalty Points that can be used to purchase ships, mods, and items that can be used or sold off.
Plexes in Faction Warfare can be found using the ship's onboard scanner and are visible to the entire system for anyone to enter... friend or foe alike.

In general high, low, and null-sec terms... "plexes" are locations that either have to be probed down or found using the ship's onboard scanner. They are basically "impromptu missions" where you have to kill NPCs, hack, salvage, and/or analyze certain objects to get useful items for sale or use. Sometimes they are linked to other plexes in other areas (ex. find an item in this plex that is needed to enter a better plex) which are commonly referred to as "escalations."
Like FW plexes... anyone can find them, but they are not announced in system and have limited to no restrictions for what ship(s) can enter them.
